What kind of enzyme is HindIII?
HindIII (pronounced “Hin D Three”) is a type II site-specific deoxyribonuclease restriction enzyme isolated from Haemophilus influenzae that cleaves the DNA palindromic sequence AAGCTT in the presence of the cofactor Mg2+ via hydrolysis.
What is the palindrome for HindIII?
The structures showed that HindIII belongs to the EcoRI-like (alpha-class) subfamily of type II restriction endonucleases. The cognate DNA-complex structures revealed the specific DNA-recognition mechanism of HindIII by which it recognizes the palindromic sequence A/AGCTT.
Is HindIII methylation sensitive?
Not sensitive to dam, dcm or mammalian CpG methylation.
Where does the enzyme HindIII cut?
Thermo Scientific HindIII restriction enzyme recognizes A^AGCTT sites and cuts best at 37°C in R buffer. See Reaction Conditions for Restriction Enzymes for a table of enzyme activity, conditions for double digestion, and heat inactivation for this and other restriction enzymes.
Does Hind 3 produce sticky ends?
Option B: Hind 3: It is a type 2 restriction endonuclease which gives sticky ends. It is isolated from Haemophilus influenzae.
Does HindIII produce sticky or blunt ends?
Recognition Sequences
Enzyme | Organism | Blunt or Sticky End |
---|---|---|
HindIII | Haemophilus influenzae Rd | Sticky |
Hinfl | Haemophilus influenzae Rf | Sticky |
Sau3A | Staphylococcus aureus | Sticky |
AluI | Arthrobacter luteus | Blunt |
What temperature does HindIII work best in and why?
What is HindIII-HF®?
More Product Information HindIII-HF® is an E. coli strain that carries the cloned and modified HindIII gene from Haemophilus influenzae Rd (ATCC 51907). High Fidelity (HF®) Restriction Enzymes have 100% activity in CutSmart® Buffer; single-buffer simplicity means more straightforward and streamlined sample processing.
What is E coli HindIII?
An E. coli strain that carries the HindIII gene from Haemophilus influenzae Rd (ATCC 51907). One unit is defined as the amount of enzyme required to digest 1 µg of λ DNA in 1 hour at 37°C in a total reaction volume of 50 µl.
