What is laminin 511?
Laminin-511 is a major component of the basement membrane, which is expressed in early development of the embryo and can be used as a matrix for pluripotent (ES/iPS) stem cells, as it binds to integrin on cell surfaces.
What is laminin 521?
Recombinant human Laminin 521 (rhLaminin-521) is a recombinant human protein that provides a defined surface for feeder-free culture of human pluripotent stem cells (PSCs).

How do you dilute Matrigel?
Matrigel should be used in µg/ml. I dilute it to a concentration of about 80µg/ml (1mg in 12ml od ice-cold DMEM-F12). This works very good with hESC H9 line. Coat dishes for at least 1h @ +37°C.
How do you inject Matrigel?
Using a 1-cc syringe and an 18-G needle, draw up 100 μL of cells and 100 μL of Matrigel. Mix the Matrigel and cells in the syringe and remove the air from the syringe. Using Matrigel should provide close to a 99% take rate. Keep your syringes on wet ice to prevent the Matrigel from solidifying prior to injection.

How much is Matrigel per well?
For endothelial tube formation, the Corning Matrigel matrix concentration should be at least 10 mg/mL. For a 24-well plate coating, we recommend 0.289 mL of chilled Matrigel matrix (10 mg/mL) per well.
How do you mix cells with Matrigel?
Make sure to resuspend your cells in such a concentration so that the desired number of cells injected in each mouse is suspended in 50 ul media. Add this to the matrigel and mix slowly by pipetting up and down ON ICE. Matrigel also gives bubble quickly, so be gentle. Inject 100 ul of this mixture to each mouse.
